Set the signal back to the boundary station and the train will set off. On some levels the train will automatically turn around if the platform is a dead end, but If you need to turn the train around simply right click it. If the train is scheduled to stop at the boundary station it will do so, then because you set the signal it will run to the next stop. They will never enter the map at a station in the middle (two signals, one at each end).
